- 體外研究:
在NIH 3T3細(xì)胞中,SU 6656抑制PDGF介導(dǎo)的S期誘導(dǎo),其IC50值為0.3-0.4 μM。在正常和集落刺激因子1受體轉(zhuǎn)染的NIH 3T3細(xì)胞中,SU 6656可抑制PDGF和血清介導(dǎo)的NIH 3T3細(xì)胞增殖及表皮生長因子和集落刺激因子1誘導(dǎo)的DNA合成。SU6656可抑制PDGF誘導(dǎo)的c-Myc感應(yīng)和ERK2活性。Jurkat T細(xì)胞經(jīng)SU 6656預(yù)處理可增加VSV-G熒光素酶活性。在近端腎小管上皮細(xì)胞中,SU 6656削弱TGF-β介導(dǎo)的CTGF mRNA和蛋白的上調(diào),也減少暴露在自分泌生長因子的細(xì)胞中的CTGF的表達(dá)。SU 6656可干擾Aurora激酶活性,造成對(duì)細(xì)胞分裂和多葉核形成的抑制
- 體內(nèi)研究:
在WSS和焦慮評(píng)分測量小鼠中,SU 6656可顯著劑量依賴性抑制mecamylamine誘導(dǎo)的實(shí)驗(yàn)性尼古丁戒斷綜合癥。SU 6656(1.5、3和6 mg/kg,腹腔注射)每日給藥一次,顯著劑量依賴性地抑制納洛酮介導(dǎo)的嗎啡戒斷綜合征
- 細(xì)胞實(shí)驗(yàn): Cell lines: HKC-8細(xì)胞 Concentrations: 5 μM Incubation Time: 48 h Method: 以不同密度(10,000 cells/cm2 and 50,000 cells/cm2)接種HKC-8細(xì)胞,經(jīng)SU 6656培養(yǎng)24 h和48 h后,測定。明敞細(xì)胞圖片經(jīng)Olympus CK40顯微鏡和Leica DC Viewer軟件拍攝
