- 體外研究:
LOXO-195在體外能有效地抑制不同的活化TRK激酶。在濃度為1 μM時(shí),LOXO-195對(duì)目標(biāo)靶點(diǎn)的選擇性比對(duì)98%其他檢測(cè)激酶(庫(kù)中包含228種激酶)高1000倍以上。在包含TRK融合的KM12、CUTO-3和O-91細(xì)胞系中,LOXO-195能有效地抑制其細(xì)胞增殖(IC50 ≤ 5 nmol/L)。而在不包含TRK融合的84種細(xì)胞系中,濃度即使高達(dá)10 μM的LOXO-195對(duì)細(xì)胞生長(zhǎng)也沒(méi)有抑制作用

在由ΔTRKA驅(qū)動(dòng)的腫瘤中,LOXO-195有效地減少磷酸化TRKA水平。在四種依賴于TRKA的腫瘤模型中(通過(guò)接種包含NIH 3T3 ΔTRKA, ΔTRKA G595R, ΔTRKA G667C和TPM3-NTRK1融合的KM12細(xì)胞構(gòu)建的腫瘤模型),LOXO-195可引起腫瘤生長(zhǎng)抑制作用
